
Acetyl Tetrapeptide-5, also marketed under the trade name Eyeseryl®, is a naturally derived tetrapeptide widely recognized for its effectiveness in reducing under-eye puffiness and dark circles while helping prevent their recurrence.
Periorbital puffiness commonly develops due to fluid accumulation in the delicate eyelid tissues, a condition known as eyelid edema. This fluid retention is often linked to impaired lymphatic drainage and increased capillary permeability, which allow excess fluid to leak into surrounding tissues.
Another contributing factor to under-eye bags is glycation, a non-enzymatic chemical process in which sugars bind to proteins. During glycation, the aldehyde group of glucose or other monosaccharides reacts with amino groups on proteins to form unstable compounds that evolve into more permanent structures known as advanced glycation end products (AGEs). These reactions promote protein cross-linking, particularly within collagen fibers, leading to reduced elasticity, tissue stiffness, and the visible formation of under-eye swelling.

This enzyme plays a vital role in protecting cells from oxidative stress by converting harmful superoxide radicals into oxygen and hydrogen peroxide. When SOD undergoes glycation, its activity becomes impaired. Research evaluating Eyeseryl® demonstrated its capacity to prevent fructose-induced glycation of SOD, resulting in preserved enzymatic activity and improved antioxidant defense.
By inhibiting protein glycation and reducing vascular leakage, Acetyl Tetrapeptide-5 helps decrease fluid accumulation while strengthening the structural integrity of the under-eye area. In addition, it enhances skin elasticity, smoothness, and provides a visible decongesting effect.
